HPLC-UV

UV detection

Ultraviolet (UV) detectors measure the ability of chromophores in compounds in a sample to absorb UV light (190-400 nm). They are located immediately after the stationary phase of the LC column to detect compounds as they elute from the column.

The majority of organic compounds can be analysed by UV detectors. UV detection is the most usual and widespread detection principle in HPLC.

The analytical development department is equiped with:

  • Variable wavelength detectors: individual wavelenghts are measured sequentially.
  • Diode array detectors: a spectrum of wavelenghts are measured simultaneously.

In both cases, the presence of an analyte gives a response assumed to be proportional to the concentration. The instrument’s response to the analyte in the unknown should be compared with the response to a standard.
